Instructions for culturing
  1. one bag of materials PER PERSON
  2. one petri dish to be kept cold (fridge; 4-7 *C)
  3. one petri dish to be kept at room temperature (around 21 *C)
  4. divide each petri dish in two halves: A and B
  5. write warm on one petri dish and cold on the other
  6. when writing: WRITE ON THE BOTTOM (not on the lid)
  7. choose two locations to swab: A and B
  8. collect the swabs and use the q-tips to infect the dishes
  9. collect pictures and data for at least two weeks

When swabbing...
  1. keep the dishes closed as long as you can.
  2. move the q-tip over the object that you want to sample
  3. open the lid and gently WITHOUT PRESSURE stroke the q-tip over the agar surface (otherwise the agar will tear)
  4. remember: A and B are separate locations!
  5. quickly close the lid
  6. when swabbing the agar, keep the plate up-side-down

TIPS
- Don't wait too long with swabbing: the quality of the agar deteriorates 
- NEVER open the lid when bacterial cultures have appeared.

  3. find a seat next to each other and re-read the following parts 1, 2 and 3 of the project introduction (YR3_PROJECT_CH10)
  4. You also find the YR3_PROJECT_CH10 document in the 'labreport: hand in your first draft here'-assignment. Divide roles in this document in part 2 of the document. 
  5. Write down your research question and hypothesis in part 3 of the document. 
  6. Discuss and determine your locations (A and B)
  7. Start writing Chapters 1 and 2 of the lab report in the 'first draft project CH10' document
  8. Are there any remaining questions?
