DNA extraction
Answer the questions on the
instruction sheet

We will work in the lab so
we follow lab rules!

Slide 4 - Slide

rules - lab experiments
  • zero-tolerance : misbehave ==> 201
  • act WOLF (Weloverwogen / Oplettend/
            Leergierig / Fijn)
  • NO eating or drinking
  • NO running , pushing or throwing

  • NO yelling
  • DON'T touch materials you don't need
  • respect each other and the materials!!!

Slide 5 - Slide

What will we do?
  1. Start with answering the questions so you know what you are doing
  2. Prepare the extraction solution water + soap + salt
  3. To extract: separate the DNA from the strawberry cells
  4. Precipitate ("neerslaan"): cause (a substance) to be deposited in solid form from a solution
  5. Clean up!!!

Slide 6 - Slide

What will we do? Tips!
- Add the soap to the beaker!!
- Add the extraction solution after the "mash and smash"!

Remember the names?
Soap

Let's get to work!
Bring:
pen/pencil

Good luck!

You only have 40 minutes!!!

Follow up DNA extraction
What did we do?

Why did we add salt?
Why did we add soap/detergent?

Why did we add alcohol?

Slide 9 - Slide

Salt: Using salt helps eliminate the histones, proteins around which DNA is wrapped.
Soap/detergent: The detergent destroys the membrane of the cell and the nucleus, allowing the DNA to come out
Alcohol: adding an ice-cold alcohol knocks DNA out of solution. This is because DNA is less soluble in an alcohol than in water.

  • BS2 -> weefsels / orgaan / functie
  • BS3 + 4 -> herken organellen naam + functie / verschil cel dierlijk + planten

wedstrijdje in duo's eukaryote cel
  1. DNA - > transcriptie naar RNA
  2. RNA -> translatie naar eiwit
eiwit is in dit geval een zin

het duo met de eerste 4 zinnen juist heeft wint

kan niet tegelijkertijd!!
help je buren niet te veel!!
